Factor XII (Hageman Factor), human (frozen)
Factor XII (FXII, Hageman Factor) is a single chain zymogen, Mr of 80,000 that circulates in plasma at a concentration of 40 g/mL and is a key component in the initiation of the intrinsic coagulation pathway. FXII binds to anionic surfaces, undergoes a conformation change, and is converted by Kallikrein to FXIIa (2-chain, alpha form). This bound alpha-form of FXIIa converts Factor XI to XIa.
Kallikrein cleaves bound FXIIa at a secondary site beyond the disulfide bond to generate a beta form of FXIIa that consists of the COOH-terminal light chain, Mr of 28,000, which contains the catalytic domain. This beta form does not bind to anionic surfaces, does not possess procoagulant activity, but does activate prekallikrein to Kallikrein.
Factor XII is purified from fresh frozen plasma by immunoaffinity.
